Yashtimadhu is a single-herb formulation prepared from the root extract of Yashtimadhu (Glycyrrhiza glabra). Yashtimadhu has demulcent and emollient actions and is recommended in the management of acid peptic disorders including non-ulcer dyspepsia. Yashtimadhu promotes the healing of gastric and duodenal ulcers by reducing basal and histamine-induced gastric acid secretion. Yashtimadhu consists of glabridin and glabrene, which exhibit inhibitory activity against the growth of Helicobacter pylori. Glycyrrhizin (as carbenoxolone sodium) increases pH at the stomach antrum, reduces reflux, improves the way food gets through to the stomach, and protects the walls of the esophagus. Yashtimadhu has a soothing property and is used as demulcent-it acts on the mucosal layer corticoster and increases the production of mucus, thus accelerating the healing of gastric and duodenal ulcers.
Pharmacological Actions
1. Gastric secretion-inhibitory activity
Glycyrrhizin (as carbenoxolone sodium) obtained from Yashtimadhu speeds the healing of gastric ulcers and protects the gastric mucosa by increasing the pH at the stomach antrum from 1.1 to 6.0 and reduces basal and histamine-induced gastric acid secretion.
2. Mucoprotective activity
Deglycyrrhizinated licorice, a derivative of glycyrrhizic acid (GA), a component of Yashtimadhu is used in the treatment of ulcers as it promotes healing by increasing mucus production and blood supply to the damaged stomach mucosa.
3. Helicobacter pylori-inhibitory activity
Yashtimadhu consists of chemical constituents such as glabridin and glabrene, which exhibit inhibitory activity against the growth of H pylori. These flavonoids also exhibit the anti-H pylori activity against a clarithromycin-resistant and amoxicillin-resistant strain.2 Orally administered glycyrrhizin is converted into the active metabolite GA by human intestinal bacteria through B-glucuronidase. GA is found to be the most active constituent in inhibiting H pylori.
4. Antireflux activity
Yashtimadhu is used to treat GERD because of its soothing property. Yashtimadhu reduces reflux by protecting the mucosal layers from the harmful effects of stomach acid, improving the way food gets through to the stomach, and protecting the walls of the esophagus.
5. Demulcent activity
Yashtimadhu has a soothing property and is used as a demulcent to promote the healing of gastric and duodenal ulcers. It acts on the mucosal layer and increases the production of mucus, thus accelerating the healing of gastric and duodenal ulcers.
